MIC135 is a modified immunocytokine (MIC) designed for the treatment of acute myeloid leukemia (AML). It consists of an Fc-optimized monoclonal antibody targeting CD135 (also known as Fms-like tyrosine kinase 3 or FLT3) fused to a mutated version of interleukin-15 (IL-15). The IL-15 component is engineered with abolished binding to the IL-15 receptor alpha (IL-15Rα), a modification intended to prevent non-specific systemic immune activation and reduce off-target toxicity. The therapeutic mechanism relies on the antibody portion to tether the cytokine to CD135-expressing leukemic cells, where it can then trans-present the IL-15 to stimulate the IL-15 receptor beta/gamma (IL-15Rβγ) on natural killer (NK) cells. This localized activation induces target-restricted NK cell proliferation and enhances antibody-dependent cellular cytotoxicity (ADCC) against AML cells while sparing healthy tissues.
