Overview

Microbial Transplantation Therapy (MTT) is an investigational live biotherapeutic product developed by Boston Children's Hospital, in collaboration with the University of Minnesota, for the treatment of peanut allergy. The therapy involves the oral administration of encapsulated fecal microbiota derived from rigorously screened, healthy, non-allergic donors. The primary objective is to restore a diverse and healthy gut microbiome, which in turn modulates the host's immune system to promote immune tolerance toward peanut allergens. While early studies utilized frozen capsules of donor stool, the refined MTT version contains concentrated bacteria with less than 1% fecal material, allowing for easier storage and home administration. Clinical results from a Phase 1 trial (NCT02960074) led by Dr. Rima Rachid demonstrated that a single treatment could significantly increase peanut tolerance in approximately 40% of participants, with effects lasting several months. A Phase 2 trial (NCT05695261) is currently evaluating the efficacy and safety of the purified MTT formulation in pediatric patients.
