Overview

microRNA-100 (miR-100) is a naturally occurring microRNA that acts as a tumor suppressor in various malignancies, including glioblastoma multiforme (GBM). It functions by post-transcriptionally regulating gene expression, specifically targeting the Silencing Mediator for Retinoid and Thyroid Hormone Receptors (SMRT), also known as Nuclear Receptor Corepressor 2 (NCOR2). In glioblastoma, miR-100 expression is typically downregulated compared to normal brain tissue. Preclinical research has demonstrated that the delivery of miR-100 precursors or mimics can inhibit tumor cell proliferation, induce apoptosis, and significantly extend survival in intracranial xenograft models. These effects are mediated through the downregulation of SMRT, which in turn reduces the Ki-67 proliferation index and triggers programmed cell death.
