microRNA-125a (miR-125a) is a highly conserved microRNA that functions as a tumor suppressor across various human malignancies, including cervical, breast, and hepatocellular carcinoma. As a therapeutic candidate, miR-125a is typically developed as a synthetic microRNA mimic designed to restore the function of the endogenous miRNA, which is frequently downregulated in malignant tissues. Its primary mechanism of action involves the post-transcriptional silencing of oncogenic targets by binding to their 3' untranslated regions (UTRs). A key target is the Signal Transducer and Activator of Transcription 3 (STAT3); by downregulating STAT3, miR-125a inhibits cell proliferation, induces apoptosis, and reverses chemoresistance to agents such as paclitaxel and cisplatin. Additionally, miR-125a has been shown to target the ERBB family (HER2 and HER3), further contributing to its anti-tumor activity. While currently in the preclinical stage of development, miR-125a represents a promising strategy for targeted RNA therapy in treatment-resistant malignancies.
