microRNA-1293 (miR-1293) is a tumor-suppressive microRNA being investigated for the treatment of diffuse pleural mesothelioma (DPM), particularly in cases characterized by NF2 loss or Hippo pathway disruption. It functions by targeting the S100A16-centered Hippo/EMT axis, leading to the downregulation of S100 calcium binding protein A16 (S100A16), YAP, TAZ, and TEAD, as well as markers of epithelial-mesenchymal transition (EMT). miR-1293 re-expression induces apoptosis and cell-cycle arrest in mesothelioma cells. For therapeutic delivery, it has been formulated in a silk fibroin hydrogel (SFH-miR-1293) for local administration, which has demonstrated significant anti-tumor activity and survival benefits in preclinical orthotopic models.
