microRNA-148a mimic is a synthetic double-stranded RNA oligonucleotide designed to replicate the biological activity of the endogenous tumor suppressor microRNA-148a (miR-148a). miR-148a is frequently downregulated in various malignancies, including renal cell carcinoma (RCC), gastric cancer, and hepatocellular carcinoma. The mimic acts by binding to the 3' untranslated regions (3'-UTRs) of specific target oncogenic mRNAs, leading to their degradation or translational repression. Key validated targets include WNT10a, which modulates the Wnt/beta-catenin signaling pathway, and DNA methyltransferase 1 (DNMT1), which is involved in epigenetic gene silencing. In preclinical models of RCC, miR-148a mimics have been shown to inhibit tumor cell proliferation, migration, and invasion, while also enhancing sensitivity to therapeutic agents such as arsenic trioxide. Although primarily used as a research tool, miR-148a mimics are being explored as potential RNA-based therapeutic candidates for cancer treatment.
