microRNA-98-5p mimic is a synthetic double-stranded RNA oligonucleotide designed to emulate the biological activity of the endogenous tumor-suppressive microRNA, miR-98-5p. In non-small cell lung cancer (NSCLC), miR-98-5p expression is frequently downregulated, leading to the overexpression of its oncogenic targets. The mimic functions by binding to the 3'-untranslated region (UTR) of specific messenger RNAs, notably mitogen-activated protein kinase kinase kinase kinase 3 (MAP4K3) and insulin-like growth factor 2 (IGF2), thereby inducing their degradation or inhibiting their translation. Downregulation of MAP4K3 subsequently suppresses the mTOR signaling pathway, resulting in inhibited cell proliferation and induced apoptosis. Additionally, the mimic can restore sensitivity to chemotherapeutic agents like paclitaxel by counteracting the effects of circular RNAs (such as hsa_circ_0003489) that sponge endogenous miR-98-5p.
