Micvotabart pelidotin is an investigational, first-in-concept **antibody-drug conjugate** developed by Pyxis Oncology. It uses a fully human IgG1 antibody to bind the extra-domain B splice variant of fibronectin in the tumor extracellular matrix, enabling extracellular protease-mediated release of an Auristatin-0101 cytotoxic payload. The released payload disrupts microtubule formation and can diffuse locally to produce direct tumor-cell killing and bystander activity; tumor-cell death is also intended to promote immunogenic cell death and remodeling of the tumor microenvironment. Micvotabart pelidotin is being evaluated as monotherapy and with pembrolizumab in advanced solid tumors, including recurrent or metastatic head and neck squamous cell carcinoma.
