MID-00935 is a novel small molecule disruptor of the MG53-IRS1 protein-protein interaction, developed by CrystalGenomics for the treatment of metabolic disorders. MG53 (also known as TRIM72) is an E3 ubiquitin ligase that mediates the ubiquitination and subsequent degradation of the insulin receptor substrate 1 (IRS-1), a key component of the insulin signaling pathway. By blocking the interaction between MG53 and IRS-1, MID-00935 prevents IRS-1 degradation, thereby enhancing insulin-induced phosphorylation of Akt and Erk. In preclinical models of diet-induced obesity (DIO), oral administration of MID-00935 has been shown to lower fasting blood glucose levels and improve glucose disposal, suggesting its potential as a muscle-targeting therapeutic for insulin resistance, type 2 diabetes, and obesity.
