Overview

Midazolam + bupropion is a combination of two pharmaceutical drugs with distinct mechanisms of action. Midazolam is a short-acting benzodiazepine that acts as a positive allosteric modulator at the GABA(A) receptor, enhancing the inhibitory effects of gamma-aminobutyric acid (GABA) in the central nervous system. This results in sedative, anxiolytic, amnestic, muscle relaxant, and hypnotic effects[2]. Bupropion is primarily a norepinephrine-dopamine reuptake inhibitor (NDRI), weakly inhibiting the reuptake of norepinephrine and dopamine by binding to their respective transporters (NET and DAT), thereby increasing their synaptic concentrations[4][6]. Bupropion also acts as an antagonist at nicotinic acetylcholine receptors[6]. The combination does not represent an approved fixed-dose product but may be encountered clinically when both drugs are prescribed together for separate indications. Caution is advised due to potential pharmacodynamic interactions such as increased risk of CNS depression or seizures[1][2][3].
