Overview

MIDGE-TH1 is a proprietary linear DNA vector technology developed by Mologen AG. Unlike traditional circular plasmids, MIDGE (Minimalistic Immunogenically Defined Gene Expression) vectors are small, linear, double-stranded DNA molecules sealed with hairpin loops at both ends. This minimalistic design excludes unnecessary bacterial sequences, such as antibiotic resistance genes, which improves safety and reduces the risk of horizontal gene transfer. The TH1 designation indicates that the vector is specifically designed to induce a strong Th1-type immune response, characterized by the activation of cytotoxic T cells and the production of pro-inflammatory cytokines like interferon-gamma. This technology has been primarily investigated as a vaccine platform for infectious diseases, most notably leishmaniasis, and as an immunotherapeutic approach in oncology.
