Midomafetamine, commonly known as MDMA, is a synthetic entactogen being developed by Lykos Therapeutics for use in MDMA-assisted therapy. It primarily acts as a monoamine releaser, increasing the synaptic concentrations of serotonin, norepinephrine, and dopamine by binding to and reversing the direction of their respective transporters (SERT, NET, and DAT). It also acts as an agonist at several serotonin receptors, including 5-HT2A and 5-HT1A, and the trace amine-associated receptor 1 (TAAR1). In clinical settings, midomafetamine is administered in a limited number of sessions alongside intensive psychotherapy to treat conditions such as posttraumatic stress disorder (PTSD), where it is thought to facilitate emotional processing by reducing fear and increasing empathy and interpersonal trust.
