Overview

MIDRIX4 (also known as MIDRIX4-LUNG) is an autologous dendritic cell (DC) vaccine developed by Ghent University Hospital for the treatment of metastatic non-small cell lung cancer (NSCLC). The vaccine is produced by isolating a patient's own monocytes, differentiating them into dendritic cells, and loading them with four specific tumor-associated antigens (TAAs)—MAGE-A3, PRAME, NY-ESO-1, and Survivin—using mRNA electroporation. This tetravalent approach is designed to stimulate a robust and diverse T-cell response against multiple targets expressed by the tumor cells, potentially overcoming tumor heterogeneity and immune escape. The vaccine also incorporates TriMix mRNA (encoding CD40L, CD70, and caTLR4) to enhance dendritic cell maturation and potency. MIDRIX4 is currently being evaluated in a first-in-human Phase Ia clinical trial to establish its safety profile and maximal tolerated dose when administered intravenously.
