Overview

**Mifomelatide** (developmental code name **TCMCB07**) is a synthetic cyclic peptide under development as a melanocortin MC3 and MC4 receptor antagonist for the treatment of cachexia, a debilitating condition involving severe weight loss and muscle wasting often associated with cancer and other chronic diseases. Administered via subcutaneous injection, it crosses the blood-brain barrier to block MC3R and MC4R, thereby alleviating anorexia, stimulating appetite, and promoting body weight gain. Developed by Endevica Bio, mifomelatide has demonstrated safety in phase 1 trials with early efficacy signals, including robust increases in appetite and body mass, and is currently advancing in phase 2 clinical trials as of 2025, primarily targeting cancer-related cachexia.[4][6][8][12]
