MIH5 is a rat-derived monoclonal antibody (clone MIH5) that specifically binds to murine Programmed Death-Ligand 1 (PD-L1), also known as CD274. It serves as an immune checkpoint inhibitor by sterically hindering the interaction between PD-L1 and its receptor, Programmed Death-1 (PD-1). This blockade prevents the transmission of inhibitory signals to T cells, thereby promoting their activation, proliferation, and effector functions against malignant cells. MIH5 is a standard tool in preclinical immuno-oncology, used to evaluate therapeutic strategies in various murine cancer models, including melanoma, mesothelioma, and colorectal carcinoma. Research utilizing MIH5 has highlighted the importance of PD-L1 expression in tumor-draining lymph nodes for restraining systemic anti-tumor immunity.
