Overview

The **MiHA mRNA-electroporated donor dendritic cell vaccine** is an experimental, ex vivo-generated cellular immunotherapy developed by the Radboud University Nijmegen Medical Centre (Radboud UMC). It consists of donor-derived dendritic cells (DCs) that are electroporated with mRNA encoding hematopoietic-restricted minor histocompatibility antigens (MiHAs), such as HA-1 or LRH-1. Administered intravenously to patients following HLA-matched allogeneic stem cell transplantation (allo-SCT) for hematologic malignancies, the vaccine is designed to selectively boost graft-versus-tumor (GVT) immunity. By presenting these hematopoietic-restricted MiHAs, the vaccine stimulates and expands donor-derived CD8+ cytotoxic T cells to target and eliminate residual leukemia or lymphoma cells without triggering graft-versus-host disease (GvHD).
