MIIST305 is an orally delivered, gut-restricted, non-steroidal and non-biologic drug candidate developed by Synedgen for the treatment of gastrointestinal diseases. It is designed to restore mucosal healing and barrier integrity in the gut by targeting the glycocalyx and mucus layers that protect intestinal epithelial cells. MIIST305 acts at the mucosal surface to reduce activation of innate inflammatory receptors, thereby breaking cycles of inflammation and promoting tissue regeneration. The drug is being developed for two main indications: as an induction and maintenance therapy for ulcerative colitis (UC) and as a medical countermeasure for gastrointestinal acute radiation syndrome (GI-ARS). Preclinical studies have shown that MIIST305 improves intestinal epithelial barrier function, suppresses systemic inflammatory responses mediated by pro-inflammatory cytokines, restores beneficial gut microbiota diversity, and enhances survival following lethal radiation exposure in animal models. The mechanism involves modulation of innate immune receptor responses at the GI cell surface without systemic absorption[1][2][3][4][5][6].
