Overview

MIL62 + lenalidomide is a combination therapy under investigation for the treatment of relapsed or refractory indolent non-Hodgkin lymphomas, specifically follicular lymphoma (FL) and marginal zone lymphoma (MZL). MIL62 is a novel third-generation, glycoengineered anti-CD20 monoclonal antibody designed to enhance antibody-dependent cell cytotoxicity. Lenalidomide is an immunomodulatory agent that alters cytokine production, enhances T-cell co-stimulation, and increases natural killer cell-mediated cytotoxicity; it also modulates the cullin ring E3 ubiquitin ligase complex by binding to cereblon and promoting degradation of key B-cell transcription factors. The combination aims to improve efficacy in B-cell malignancies by targeting both tumor cells directly (via CD20) and modulating the immune response[3][5][6][7][8].
