Overview

Milatuzumab is a humanized monoclonal antibody that targets CD74, a cell surface molecule also known as the invariant chain of major histocompatibility complex class II. CD74 acts as a receptor for macrophage migration inhibitory factor (MIF) and plays a role in linking innate and adaptive immune responses by regulating the survival of macrophages and B cells. Milatuzumab exerts its antitumor effects primarily through antibody-dependent cell cytotoxicity and T lymphocyte stimulation. It has been investigated in clinical trials for several hematological malignancies—including multiple myeloma, chronic lymphocytic leukemia (CLL), non-Hodgkin lymphoma—as well as autoimmune diseases such as systemic lupus erythematosus (SLE) and graft-versus-host disease (GVHD). The drug was originally developed by Immunomedics (now part of Gilead Sciences). Despite promising early-phase results showing tolerability and some efficacy signals, further development has been discontinued across all indications[1][2][5].
