Overview

**Milatuzumab-doxorubicin** is an investigational CD74-directed antibody-drug conjugate composed of the humanized anti-CD74 monoclonal antibody milatuzumab covalently linked to the anthracycline payload doxorubicin. Following CD74 binding and internalization by tumor cells, doxorubicin is released and intercalates DNA while inhibiting topoisomerase II, producing replication blockade and DNA damage. The agent was initially developed by Immunomedics and subsequently studied under Gilead Sciences sponsorship in relapsed or refractory multiple myeloma, non-Hodgkin lymphoma, and chronic lymphocytic leukemia; its registered Phase I/II studies were terminated.
