Drug intelligence / Profile preview
mINDY antisense oligonucleotide is a second-generation 2'-O-methoxyethyl chimeric antisense oligonucleotide (ASO) designed to selectively knock down the expression of mammalian Indy (mINDY, also known as SLC13A5), a sodium-coupled citrate transporter primarily expressed in the liver. Developed in collaboration between Yale University and Isis Pharmaceuticals (now Ionis Pharmaceuticals), the ASO targets mINDY mRNA to reduce its expression, thereby inhibiting hepatic extracellular citrate uptake. In preclinical models of diet-induced obesity and insulin resistance, treatment with mINDY ASO significantly reduced hepatic triglyceride content, fasting plasma insulin, and plasma triglycerides, while improving hepatic insulin sensitivity and suppressing hepatic glucose production. This makes mINDY ASO a potential therapeutic candidate for the treatment of non-alcoholic fatty liver disease (NAFLD), hepatic steatosis, and type 2 diabetes.
