mini-CAR-NK CDV is an experimental, cell-free therapeutic platform consisting of nanovesicles derived from Natural Killer (NK) cells that have been genetically engineered to express a "mini" Chimeric Antigen Receptor (CAR). Unlike traditional CAR-NK cell therapies that utilize whole living cells, these cell-derived vesicles (CDVs) are produced through the mechanical extrusion or induction of engineered NK cells, resulting in nanoscale particles that retain the targeting specificity of the CAR and the innate cytotoxic machinery of the parent NK cell membrane. The "mini-CAR" construct typically utilizes a truncated receptor design, such as an scFv fused to a Glycosylphosphatidylinositol (GPI) anchor, which allows for high-density display on the vesicle surface without the need for complex intracellular signaling domains. These vesicles are designed to penetrate solid tumor microenvironments, such as glioblastoma, more effectively than whole cells while minimizing risks associated with cell therapies, such as cytokine release syndrome (CRS). Furthermore, mini-CAR-NK CDVs can be loaded with chemotherapeutic agents to provide a synergistic combination of targeted immunotherapy and localized chemotherapy.
