mini-hyperCVD is a low-intensity chemotherapy regimen primarily used for the treatment of acute lymphoblastic leukemia (ALL) and lymphoblastic lymphoma (LBL), particularly in older adults or patients unfit for intensive chemotherapy. It is a dose-reduced modification of the standard hyper-CVAD regimen. The regimen typically omits anthracyclines (such as doxorubicin) and features reduced doses of cyclophosphamide, dexamethasone, methotrexate, and cytarabine to improve tolerability while maintaining efficacy. It is structured into alternating cycles: Part A consists of cyclophosphamide, vincristine, and dexamethasone, while Part B consists of high-dose methotrexate and cytarabine. In contemporary clinical trials, mini-hyperCVD is frequently evaluated as a backbone regimen in combination with targeted agents such as venetoclax, blinatumomab, inotuzumab ozogamicin, or capivasertib.
