The miniPRO HER2 binder is a preclinical-stage therapeutic candidate being developed by Ordaos Bio. It represents a novel class of de novo designed mini-proteins (40-160 amino acids) that target the extracellular domain of the human epidermal growth factor receptor 2 (HER2). Created using Ordaos's proprietary AI-powered 'Design Engine', these miniPRO molecules are approximately 93% smaller than traditional monoclonal antibodies like trastuzumab, which potentially allows for superior penetration into tumor microenvironments. Preclinical studies have demonstrated nanomolar binding affinity (EC50 in the nanomolar range) to human HER2 and high stability, solubility, and manufacturability in mammalian protein expression systems. The drug is intended for the treatment of HER2-positive cancers, including breast, ovarian, esophageal, and cholangiocarcinoma.
