Overview

A combination therapy comprising **Minnelide** (a water-soluble prodrug of triptolide), **nab-paclitaxel** (albumin-bound paclitaxel, marketed as Abraxane), and **gemcitabine**. Minnelide acts primarily as an anti-cancer agent by inhibiting super-enhancers and the oncogene MYC, reducing HSP70 levels, and promoting apoptosis and cell cycle arrest in tumor cells. Nab-paclitaxel is a microtubule inhibitor that disrupts cell division, while gemcitabine is a nucleoside analog that inhibits DNA synthesis. This combination has shown synergistic activity in preclinical pancreatic cancer models, improving efficacy and survival, particularly in pancreatic ductal adenocarcinoma (PDAC), with research also ongoing for other GI tract malignancies. The rationale for the combination is to achieve stronger anti-tumor effects at reduced doses to minimize toxicity, targeting both tumor and tumor stroma effectively[1][3][4][5].
