Overview

Minocycline is a broad-spectrum tetracycline antibiotic used to treat a variety of bacterial infections, including respiratory tract infections, urinary tract infections, skin and soft tissue infections, severe acne, rosacea (inflammatory lesions), sexually transmitted diseases such as chlamydia and gonorrhea (especially in penicillin-allergic patients), tick-borne diseases like rickettsial infections, plague, tularemia, anthrax (including bioterrorism-related exposure), meningococcal carrier state eradication, and other zoonotic or atypical bacterial diseases. It is also sometimes used off-label for mild rheumatoid arthritis due to its anti-inflammatory properties. Mechanistically, minocycline works by binding to the 30S ribosomal subunit of bacteria and inhibiting protein synthesis by preventing aminoacyl-tRNA from attaching to the ribosome. This action is bacteriostatic rather than bactericidal. In addition to its antibacterial effects, minocycline has been shown in research settings to modulate human enzymes such as matrix metalloproteinase-9 and polyunsaturated fatty acid 5-lipoxygenase; it can also inhibit inflammatory mediators like interleukin-1 beta and nitric oxide synthase inducible isoform[5][3][7]. Minocycline was first described in 1966.
