Drug intelligence / Profile preview
Arestin (minocycline hydrochloride microspheres, 1 mg) is a locally administered, sustained-release antibiotic developed by OraPharma, a subsidiary of Bausch Health. It is composed of bioresorbable poly(glycolide-co-dl-lactide) microspheres that encapsulate minocycline, a broad-spectrum tetracycline derivative. The product is designed for subgingival administration into periodontal or peri-implant pockets, where it slowly releases the antibiotic over several days. Minocycline exerts its primary effect by binding to the 30S ribosomal subunit of bacteria, thereby inhibiting protein synthesis. Additionally, it possesses non-antibacterial properties, such as the inhibition of matrix metalloproteinases (MMPs), which are enzymes involved in the degradation of the extracellular matrix during inflammatory conditions like peri-implantitis and periodontitis. While Arestin is already approved for the treatment of chronic periodontitis, it is currently undergoing Phase 3 clinical evaluation for the treatment of peri-implantitis as an adjunct to mechanical debridement.
