Minodronate + alendronate is a combination of two bisphosphonate drugs used primarily for the treatment and prevention of osteoporosis. Both agents act by inhibiting bone resorption through suppression of osteoclast activity. Alendronate, a second-generation bisphosphonate, binds to bone hydroxyapatite and induces apoptosis in osteoclasts after being released during bone resorption[8]. Minodronate is a third-generation bisphosphonate that inhibits farnesyl pyrophosphate synthase in the mevalonic acid pathway within osteoclasts, also leading to their apoptosis[6]. Both drugs increase bone mineral density and reduce fracture risk in patients with osteoporosis. Minodronate is noted as one of the most potent oral inhibitors of bone resorption among bisphosphonates[6], while alendronate has established efficacy for postmenopausal, steroid-induced, and male osteoporosis as well as Paget’s disease[1][2][3].
