Overview

Minokitug (GRN-300) is a first-in-class monoclonal antibody developed by Granite Bio that dual-targets the inhibitory receptors **ILT2** (LILRB1) and **ILT4** (LILRB2). These receptors are key immune checkpoints expressed on myeloid cells (such as macrophages and dendritic cells) and lymphoid cells (NK and T cells) that contribute to immune evasion within the tumor microenvironment. By simultaneously blocking ILT2 and ILT4, minokitug is designed to reverse myeloid-mediated immunosuppression and promote a pro-inflammatory environment, thereby enhancing the anti-tumor activity of both the innate and adaptive immune systems. The agent is currently being evaluated in the Phase 1 **MONOlith** clinical trial for patients with advanced solid tumors.
