MIP3α-gp100 is an experimental **DNA vaccine** composed of a fusion protein linking the chemokine **Macrophage Inflammatory Protein-3 alpha (MIP3α, also known as CCL20)** with the melanoma-associated antigen **gp100**. The construct is designed to target immature dendritic cells through the MIP3α component, thereby enhancing antigen uptake and presentation by both class I and class II MHC pathways. This leads to a more robust activation of both CD4+ and CD8+ T cell responses. In preclinical models, particularly the B16F10 murine melanoma model, this DNA vaccine shows enhanced immunogenicity, reduced tumor burden, delayed tumor growth, and improved survival compared to gp100 antigen-only vaccines. Efficacy is further enhanced in combination with immune checkpoint inhibitors or IL-10 neutralization. The modular platform allows for adaption to a variety of tumor antigens and is under exploration for personalized cancer immunotherapy[1][3][4][5].
