Overview

miR-1 (microRNA-1) is a highly conserved microRNA that functions as a potent tumor suppressor in various malignancies, including small cell lung cancer (SCLC). In SCLC, miR-1 is frequently downregulated, and its therapeutic restoration is being explored to inhibit tumor growth and metastasis. The mechanism of action involves the direct targeting of the C-X-C chemokine receptor type 4 (CXCR4) mRNA, leading to its degradation or translational repression. This inhibition subsequently suppresses the FOXM1-RRM2 signaling axis, which is critical for oncogenic signaling, DNA synthesis, and cell cycle progression. Research conducted at the University of Nebraska Medical Center and City of Hope indicates that miR-1 mimics have high potential for the development of novel SCLC therapies, particularly in cases where traditional therapeutic options are limited.
