miR-10b is a microRNA that functions as a potent driver of metastasis in several aggressive cancers, including breast cancer, pancreatic adenocarcinoma, and glioblastoma. Often characterized as an "oncomiR," its overexpression promotes cell migration and invasion by silencing tumor-suppressive targets such as HOXD10 and TIP30, which in turn activates pro-metastatic pathways like RhoC. Therapeutic development primarily focuses on the inhibition of miR-10b using antisense oligonucleotides, commonly referred to as "antagomirs." The most advanced clinical-stage candidate is TransCode Therapeutics' TTX-MC138, which utilizes a magnetic iron oxide nanoparticle delivery system to transport the antagomir specifically to metastatic lesions, where it binds to and neutralizes miR-10b to inhibit further disease progression.
