miR-10b precursor hairpin inhibitor is an experimental therapeutic agent designed to selectively target the structured precursor hairpin of microRNA-10b (miR-10b). By binding to the RNA architecture of the precursor, the inhibitor disrupts the Dicer-mediated processing required for the maturation of miR-10b. miR-10b is a well-characterized oncomiR that is significantly upregulated in glioblastoma (GBM), where it drives tumor progression and invasion by silencing tumor suppressors such as PTEN and HOXD10. The inhibition of miR-10b maturation leads to the reactivation of these suppressor pathways, effectively suppressing GBM cell proliferation and invasive behavior in patient-derived models and human cerebral organoids. This approach represents a precision medicine strategy that targets noncanonical oncogenic mechanisms while minimizing effects on normal neural tissue.
