Overview

miR-124-3p antagomir is a synthetic, chemically modified antisense oligonucleotide designed to specifically bind to and inhibit the activity of the endogenous microRNA miR-124-3p. Antagomirs are typically characterized by 2'-O-methyl modifications, phosphorothioate linkages, and a 3' cholesterol conjugate, which collectively enhance metabolic stability and facilitate cellular uptake. By sequestering mature miR-124-3p, the antagomir prevents the microRNA from binding to the 3' untranslated regions (UTRs) of its target mRNAs, thereby alleviating translational repression or mRNA degradation. Preclinical research has demonstrated that miR-124-3p antagomir can modulate various pathological axes, such as the miR-124-3p/NPY axis in depression-related appetite suppression, the STAT3/NSF/GluR2 pathway in visceral hypersensitivity, and the EGR1 regulatory axis in retinal neovascularization and neuropathic pain. It is primarily utilized as a research tool to elucidate microRNA function and as a potential therapeutic candidate for neurological, inflammatory, and ocular disorders where miR-124-3p is pathologically overexpressed.
