miR-135b antagomir is an experimental antisense oligonucleotide (antagomir) designed to silence microRNA-135b (miR-135b). miR-135b is an oncomiR that is frequently upregulated in various cancers, where it promotes tumor cell proliferation, survival, and metastasis by downregulating tumor suppressor genes. A key target of miR-135b is the Large Tumor Suppressor Kinase 2 (LATS2), a core component of the Hippo signaling pathway. By inhibiting miR-135b, the antagomir restores LATS2 expression, thereby inhibiting the oncogenic activity of the YAP/TAZ complex. Preclinical studies have demonstrated that systemic or local administration of miR-135b antagomir can suppress tumor growth and enhance sensitivity to chemotherapy in models of colorectal, lung, and gastric cancer. It is also being explored for its potential to mitigate lung injury in conditions like bronchopulmonary dysplasia.
