Overview

miR-139 is a microRNA that acts as a tumor suppressor in various malignancies, most notably localized prostate cancer. It is frequently down-regulated in cancerous tissues, and its expression levels are inversely correlated with biochemical recurrence and metastasis. miR-139 functions by post-transcriptionally inhibiting the expression of several oncogenic factors, including Insulin-like Growth Factor 1 Receptor (IGF1R), Runt-related transcription factor 1 (RUNX1), and AXL receptor tyrosine kinase. By suppressing these targets, miR-139 inhibits the Akt signaling pathway and cyclin D1 expression while promoting the CDK inhibitor p21, leading to reduced cell proliferation, migration, and G2 cell cycle arrest. It is being explored both as a prognostic biomarker and as a potential therapeutic replacement therapy using miR-139 mimics.
