miR-144 inhibitor refers to a class of RNA-based therapeutic candidates, typically antisense oligonucleotides or antagomirs, designed to specifically bind and neutralize microRNA-144 (miR-144). In various malignancies, such as esophageal squamous cell carcinoma (ESCC), miR-144 has been identified as an oncogenic microRNA (oncomiR) that promotes tumor progression by directly targeting and downregulating tumor suppressor genes, most notably PURA (Purine-rich element-binding protein A). By sequestering miR-144, these inhibitors restore the expression of target genes, thereby suppressing cell proliferation, migration, and colony formation. While currently utilized primarily in preclinical research to validate miR-144 as a viable therapeutic target, miR-144 inhibitors represent a potential targeted therapy for cancers characterized by miR-144 overexpression.
