miR-145-MNPF is a magnetic nanoparticle (MNP) based nanoformulation designed for the intracellular delivery and sustained release of microRNA-145 (miR-145), a known tumor suppressor. Developed by researchers at the University of Tennessee Health Science Center, the formulation utilizes positively charged polyethyleneimine (PEI) to enhance the loading efficiency of miR-145 mimics onto the magnetic nanoparticles. In pancreatic cancer models, miR-145-MNPF restores miR-145 levels, which are typically suppressed during the progression from pancreatic intraepithelial neoplasia (PanIN) to pancreatic ductal adenocarcinoma (PDAC). This restoration leads to the significant downregulation of several oncogenic targets, including the transmembrane mucin MUC13, HER2, P-AKT, and p53. Preclinical studies indicate that miR-145-MNPF inhibits cell proliferation, colony formation, migration, and invasion, and sensitizes pancreatic cancer cells to paclitaxel and TRAIL therapy.
