Overview

**miR-146a inhibitor** refers to a chemically-modified synthetic oligonucleotide (often called an "antagomir" or "inhibitor") designed to specifically bind and inhibit the activity of endogenous microRNA-146a (miR-146a). miR-146a is a non-coding RNA involved in regulating the inflammatory response, cardiac remodeling, immune function, and more, largely through its targeting of key genes such as IRAK1 and TRAF6 and subsequent modulation of NF-κB signaling pathways. miR-146a inhibitors have been used experimentally to upregulate the expression of these target genes and increase inflammatory and fibrotic responses. In animal models, administration of miR-146a inhibitors (antagomirs) attenuated heart failure by improving cardiac function and reducing cardiac remodeling following myocardial infarction. Such inhibitors are still experimental, mostly used in preclinical studies to explore the therapeutic potential of modulating miR-146a activity for diseases such as heart failure, cardiac fibrosis, and possibly other pathological conditions related to miR-146a dysregulation. miR-146a inhibitors function via sequence-specific binding, preventing the microRNA from exerting its gene-silencing effects.
