miR-148a-3p mimic is a synthetic double-stranded RNA oligonucleotide designed to emulate the biological activity of the endogenous microRNA hsa-miR-148a-3p. In the context of multiple myeloma (MM) research, miR-148a-3p has been identified as an oncomiR that is significantly overexpressed in patients and associated with increased tumor cell proliferation. The mimic is primarily utilized as a research tool in gain-of-function studies to demonstrate that elevated levels of miR-148a-3p lead to the post-transcriptional silencing of tumor suppressor genes, including CCDC6, PTEN, SH2B3, and PREX1. By binding to the 3' untranslated regions (UTRs) of these target mRNAs, the mimic induces translational inhibition or mRNA degradation, thereby promoting oncogenic signaling pathways and cellular proliferation. While the mimic serves to elucidate disease mechanisms, therapeutic strategies in MM typically focus on miR-148a-3p inhibitors to counteract these effects.
