miR-150 is a microRNA that acts as a key regulator of hematopoietic lineage differentiation. In myeloid leukemias, including acute myeloid leukemia (AML) and chronic myeloid leukemia (CML), miR-150 expression is frequently downregulated, which contributes to the characteristic block in myeloid cell maturation. Therapeutic strategies involving miR-150 aim to restore its levels to induce differentiation and inhibit leukemic cell growth. The primary mechanism of action for miR-150 is the post-transcriptional repression of the MYB transcription factor, a protein essential for maintaining the proliferative state of hematopoietic progenitor cells. By binding to the 3-UTR of MYB mRNA, miR-150 reduces its expression, thereby promoting the transition from a progenitor state to a differentiated myeloid cell.
