Overview

miR-155-5p inhibitors are a class of RNA-based therapeutics, typically antisense oligonucleotides, designed to bind and sequester microRNA-155-5p. miR-155 is a well-characterized microRNA that plays a central role in regulating inflammatory and immune responses. In the context of diabetic complications, miR-155-5p is upregulated and acts to suppress the expression of Suppressor of Cytokine Signaling 1 (SOCS1). This suppression leads to the overactivation of the JAK/STAT signaling pathway, which promotes renal inflammation, fibrosis, and vascular damage. Therapeutic inhibition of miR-155-5p aims to restore SOCS1 levels, thereby dampening the JAK/STAT-mediated inflammatory cascade. Preclinical studies have demonstrated that miR-155-5p antagonism can alleviate albuminuria, reduce atherosclerosis burden, and improve plaque stability in diabetic models, highlighting its potential as a treatment for chronic complications of diabetes.
