miR-183C antagomir is a synthetic antisense oligonucleotide designed to inhibit the miR-183-96-182 (miR-183C) microRNA cluster. This cluster is highly conserved and has been found to be significantly upregulated in the splenic cells of murine models of systemic lupus erythematosus (SLE). By antagonizing these microRNAs, the agent increases the expression of forkhead box O1 (Foxo1), a direct target of the cluster, which in turn suppresses the production of inflammatory cytokines such as IFN-γ and IL-6. Preclinical research in B6-lpr mice has demonstrated that inhibition or depletion of miR-183C reduces anti-dsDNA autoantibody production and IgG deposition in the kidneys, suggesting its potential as a therapeutic strategy for autoimmune diseases like SLE.
