miR-196b antagomir + lactoferrin-lipid nano-assemblies is an experimental therapeutic designed to treat renal cell carcinoma (RCC) by targeting and remodeling cancer-associated fibroblasts (CAFs). The formulation consists of ultrasmall (5-7 nm) lipid nano-assemblies stabilized by recombinant human lactoferrin (rhLF). These assemblies are co-loaded with an antagomir against the oncomiR miR-196b and a small molecule inhibitor of COUP-TFII (CIA-1). The lactoferrin component serves as a targeting ligand for LRP1 and intelectin-1 receptors, which are overexpressed in fibrotic tumor tissues. By inhibiting miR-196b and COUP-TFII, the drug aims to reprogram immunosuppressive CAFs into a quiescent phenotype, thereby reducing the secretion of pro-inflammatory cytokines (IL-6, IL-8, MCP1) and enhancing the efficacy of cancer immunotherapy.
