miR-1Zip is a microRNA knockdown tool, specifically a lentiviral-based "miRZip" construct designed to permanently inhibit microRNA-1 (miR-1) activity. It functions as a microRNA sponge or decoy, expressing a short hairpin RNA that is processed into a competitive inhibitor which binds and sequesters mature miR-1. In oncological research, particularly in small cell lung cancer (SCLC), miR-1Zip is used to study the effects of miR-1 loss-of-function. Research indicates that inhibiting miR-1 with miR-1Zip leads to increased tumorigenesis and metastasis by derepressing the CXCR4/FOXM1/RRM2 signaling axis, confirming miR-1's role as a tumor suppressor.
