miR-20a inhibitors are a class of RNA-based molecules, typically antisense oligonucleotides or antagomirs, designed to selectively bind and neutralize microRNA-20a (miR-20a). miR-20a is a member of the miR-17-92 cluster, an 'oncomir' cluster that is frequently overexpressed in various malignancies, including multiple myeloma, lymphomas, and solid tumors. By sequestering miR-20a, these inhibitors prevent it from downregulating its target tumor-suppressor mRNAs, thereby promoting apoptosis and inhibiting the proliferation of cancer cells. In research settings, miR-20a inhibitors have been shown to sensitize multiple myeloma cells to proteasome inhibitors like bortezomib. While primarily used as laboratory tools to investigate microRNA function, they represent a potential therapeutic strategy for diseases characterized by miR-20a dysregulation.
