miR-216a adenovirus is a viral vector-based research tool used to overexpress microRNA-216a (miR-216a) in experimental models. In the context of cardiovascular research, miR-216a has been identified as a pro-inflammatory factor that promotes atherosclerosis progression by inducing M1 macrophage polarization. Mechanistically, miR-216a targets and inhibits the expression of Smad3 and IκBα, which subsequently activates NF-κB-responsive pro-inflammatory genes such as monocyte chemotactic protein-1 (MCP-1) and tumor necrosis factor alpha (TNF-α). This adenovirus is primarily utilized as a laboratory reagent to study the molecular pathways of plaque instability, lipid accumulation, and macrophage polarization in ApoE-/- mouse models.
