miR-218-5p agomir is a chemically modified, double-stranded microRNA (miRNA) mimic designed to enhance the activity of endogenous miR-218-5p. In preclinical research focused on Parkinson's disease, it has been shown to regulate microglia-mediated neuroinflammation and exert a protective effect on dopaminergic neurons. The mechanism of action involves the direct targeting and downregulation of DEAD-box helicase 41 (DDX41), which subsequently inhibits the Type I interferon (IFN-I) response pathway. By suppressing the expression of IFN-I related genes such as Irf7, Ddx60, and Nlrc5, the agomir reduces the induction of inflammatory cytokines and alleviates motor dysfunction in animal models.
