miR-219 is a microRNA that serves as a critical regulator of oligodendrocyte differentiation and myelination within the central nervous system. It functions by post-transcriptionally repressing several inhibitory factors of myelination, including Platelet-derived growth factor receptor alpha (PDGFRα) and Forkhead box J3 (Foxj3). In the context of vascular dementia (VaD) and other white matter diseases, miR-219 expression is often reduced, leading to impaired oligodendrocyte progenitor cell (OPC) maturation and subsequent white matter damage. Therapeutic approaches focusing on the augmentation of miR-219 levels, such as through the use of synthetic miR-219 mimics or gene therapy, are being investigated to promote remyelination, restore white matter integrity, and alleviate cognitive and behavioral deficits associated with vascular dementia and multiple sclerosis.
