Overview

miR-219 mimic is a synthetic microRNA designed to replicate the biological activity of endogenous miR-219, a critical regulator of oligodendrocyte differentiation and myelin sheath assembly. In preclinical models of neuroinflammatory and demyelinating conditions, such as Theiler's murine encephalomyelitis virus-induced demyelinating disease (TMEV-IDD), intranasal administration of the mimic has been shown to ameliorate clinical symptoms, reduce neurogliosis, and promote motor and sensorimotor function recovery. The therapeutic effect is mediated through the negative regulation of proinflammatory cytokines and the inhibition of viral RNA replication. Furthermore, the mimic downregulates essential genes in the cholesterol biosynthesis pathway, including Cyp51 (lanosterol 14-alpha-demethylase) and Srebf1 (sterol regulatory element-binding protein-1), which are critical for the metabolic processes underlying myelin repair.
